Newts' Ability to Regenerate Tissue Replicated in Mouse Cells(viewbestads.com)

 


ScienceDaily — Tissue regeneration a la salamanders and newts seems like it should be the stuff of science fiction. But it happens routinely. Why can't we mammals just re-grow a limb or churn out a few new heart muscle cells as needed? New research suggests there might be a very good reason: Restricting our cells' ability to pop in and out of the cell cycle at will -- a prerequisite for the cell division necessary to make new tissue -- reduces the chances that they'll run amok and form potentially deadly cancers.
Now scientists at the Stanford University School of Medicine have taken a big step toward being able to confer this regenerative capacity on mammalian muscle cells; they accomplished this feat in experiments with laboratory mice in which they blocked the expression of just two tumor-suppressing proteins. The finding may move us closer to future regenerative therapies in humans -- surprisingly, by sending us shimmying back down the evolutionary tree.

"Newts regenerate tissues very effectively," said Helen Blau, PhD, the Donald E. and Delia B. Baxter Professor and a member of Stanford's Institute for Stem Cell Biology and Regenerative Medicine. "In contrast, mammals are pathetic. We can regenerate our livers, and that's about it. Until now it's been a mystery as to how they do it."

Blau is the senior author of the research, which will be published in Cell Stem Cell on Aug. 6. Kostandin Pajcini, PhD, a former graduate student, and Jason Pomerantz, MD, a former postdoctoral scholar in Blau's laboratory, are primarily responsible for the work and are first author and co-senior author, respectively.

Although there's been a lot of discussion about using adult or embryonic stem cells to repair or revitalize tissues throughout the body, in this case the researchers weren't studying stem cells. Instead they were investigating whether myocytes, run-of-the mill muscle cells that normally don't divide, can be induced to re-enter the cell cycle and begin proliferating. This is important because most specialized, or differentiated, cells in mammals are locked into a steady state that does not allow cell division. And without cell division, it is not possible to get regeneration.

In contrast, the cells of some types of amphibians are able to replace lost or damaged tissue by entering the cell cycle to give rise to more muscle cells. While doing so, the cells maintain their muscle identity, which prevents them from straying from the beaten path and becoming other, less useful cell types.

Pomerantz and Blau wondered if it could be possible to coax mammalian cells to follow a similar path. To do so, though, they needed to pinpoint what was different between mammalian and salamander cells when it comes to cell cycle control. One aspect involves a class of proteins called tumor suppressors that block inappropriate cell division.

Previous research had shown that a tumor suppressor called retinoblastoma, or Rb, plays an important role in preventing many types of specialized mammalian cells, including those found in muscle, from dividing willy-nilly. But the effect of blocking the expression of Rb in mammalian cells has been inconsistent: In some cases it has allowed the cells to hop back into the cell cycle; in others, it hasn't.

The researchers employed some evolutionary detective work to figure out that another tumor suppressor called ARF might be involved. Like Rb, ARF works to throw the brakes on the cell cycle in response to internal signals. An examination of the evolutionary tree provided a key clue. They saw that ARF first arose in chickens. It is found in other birds and mammals, but not in animals like salamanders nestled on the lower branches. Tellingly, it's also missing in cell lines that begin cycling when Rb is lost, and it is expressed at lower-than-normal levels in mammalian livers -- the only organ that we humans can regenerate.

Based on previous investigators' work with newts, Blau said it "seemed to us that they don't have the same limitations on growth. We hypothesized that maybe, during evolution, humans gained a tumor suppressor not present in lower animals at the expense of regeneration."

Sure enough, Pajcini and Pomerantz found that blocking the expression of both Rb and ARF allowed individual myocytes isolated from mouse muscle to dedifferentiate and begin dividing. When they put the cells back into the mice, they were able to merge with existing muscle fibers -- as long as Rb expression was restored. Without Rb the transplanted cells proliferated excessively and disrupted the structure of the original muscle.

"These myocytes have reached the point of no return," said Blau. "They can't just start dividing again. But here we show that temporarily blocking the expression of just two proteins can restore an ancient ability to contribute to mammalian muscle."

The key word here is "temporarily." As is clear from the mouse experiments, blocking the expression of tumor suppressors in mammalian cells can be a tricky gambit. Permanently removing these proteins can lead to uncontrolled cell division. But, a temporary and well-controlled loss -- as the researchers devised here -- could be a useful therapeutic tool.

The research required some sophisticated technology to separate individual myocytes from one another for study. To do so, Pajcini traveled to Munich to learn how to optimize a technique normally used on cryopreserved and fixed tissue sections -- "laser micro-dissection catapulting" -- for use with living cells. But the effort paid off when he was able to prove conclusively that once the expression of the two proteins was blocked, individual live cells were, in fact, dividing in culture.

Next, the researchers would like to see if the technique works in other cell types, like those of the pancreas or the heart, and whether they can induce it to happen in tissue at sites of injury. If so, it may be possible to trigger temporary cell proliferation as a means of therapy for a variety of ailments.

In addition to Blau, Pajcini and Pomerantz, other Stanford researchers involved in the study include senior research scientist Stephane Corbel, PhD, and assistant professor of pediatrics and genetics Julien Sage, PhD. Pajcini is now at the University of Pennsylvania, and Pomerantz is an assistant professor of surgery at the University of California-San Francisco.

Increase Your Recent Posts and Comments to 10 – Blogger Hack

How to Increase Your Post Limit

There’s a cool way to increase the number of recent posts showing up in your rss feed widget. All you need to do is add the parameter “?max-results=10” (or whatever number you want to return is) to the end of your existing feed url. So your new feed widget url should look something like this:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/?max-results=10
Pretty cool huh? Now it might not work right away in your feed widget so to make sure it’s working properly is to view it in your browser. Yup, just put it right into your browser window and you’ll see if it’s working properly because trying to troubleshoot it after it’s in your rss feed widget is really tough — especially since it doesn’t always update right away.

Filter Your Post List by Category (Tag, Label)

Say you only want to show a certain list of posts based on category in your rss feed widget. Well, now you can by using the “category” parameter. Here’s what your url would look like:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/-/cure/
I was only able to get this to work with one category but according to the example below, you can filter by multiple categories. If any of you are able to get that to work, please let me know.

Filter Your Post List by a Date Range

If you wanted to restrict your feed results to a certain time frame you can. I can’t think of a good reason for me to personally read this since I always want the latest posts to be listed but the filter is here to use regardless. Here’s an example which will only show posts that were published between 1/16/2008 – 2/28/2008:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/?published-min=2008-01-
16T00:00:00&published-max=2008-02-28T23:59:59

You can also substitute the published-min and published-max with updated-min and updated-max if you wanted to filter by updated dates instead of published dates.

Order By Filter

I actually don’t ever modify this since the default order by filter is lastmodified or createdate which makes the most recent post appear first on the list. If you wanted to change this for some reason you easily could by using the “orderby” filter.

Start Index Filter

This one could be handy if you wanted to skip certain posts and start listing after, say the 1st post. Maybe you have a section where you feature the latest post on your blog and you don’t want it to also show up first in your rss feed. This is the filter you would use. An example of the “start-index” filter would be:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/?start-index=2
This would essentially skip and not show your latest post and post #2 would come up first. Be careful if you do this and later wonder why your first post isn’t show up.

Multiple Parameter Example

You can combine all these filters into one url and really fine tune your results if you wanted to. Let’s say you want to show your 10 most recent posts filtered by the “book” category (label, tag). Your feed url would look something like this:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/-/cure/?max-results=10
Another example would be to show 10 most recent posts filtered by a date range:
http://YOUR-BLOG-NAME-HERE.blogspot.com/feeds/posts/default/?published-min=2008-01-
16T00:00:00&published-max=2008-02-28T23:59:59&
max-results=10

Here’s a list of all the query parameters you can change for any Blogger rss feed. I wouldn’t worry about the “alt” one but the rest are pretty useful to know.

  • alt – the type of feed to return, such as atom (the default) or rss.
  • orderby – the order to return entries, such as lastmodified (the default) and starttime.
  • max-results – the maximum number of entries to return.
  • /category – specifies categories (i.e. labels) to filter the feed results. For example, http://www.blogger.com/feeds/blogID/posts/default/-/Fritz/Laurie will return entries labeled with Fritz AND Laurie.
  • updated-min, updated-max – the bounds on entry update dates.
  • published-min, published-max – the bounds on entry publication dates.
  • start-index – 1-based index of the first result to be retrieved (for paging).
Now that you are familiar with all your rss feed url options, you can really customize your Blogger recent posts and comments sections. Many of you have asked me how to increase your feed limit from 5 to 10 so now you can. In some cases I had to completely delete my recent comments widget and install it again for the filters to correctly work. I’m not sure why but you might run into a similar issue.

Opera Mini as a PC Browser (big screen)(java4me.blogspot.com+bizztech.com)

METHOD-1

a new version of OperaMiniLargeDevice (OMLD) for the new Java Platform Micro Edition Software Development Kit 3.0, which is compatible with the new Opera Mini 5, has been published here. OMLD for the older Sun WTK, which is the topic of this post, will stay online, as it is also compatible with Linux and many users have installed this solution already.

Now the original post:

Today I have worked on a solution to make the famous mobile browser Opera Mini run in a big window on the PC. The reason for this is the fast browsing experience on mobile devices with this application and the small amount of traffic it generates during browsing sessions, because the Opera Mini proxy reduces page sizes. So also PC users could benefit from using this browser, especially from version 4 that introduces fullscreen support.

Because Opera Mini is a Java ME application, an emulator is necessary. Therefore, I used the Sun Java Wireless Toolkit with the included MIDP emulator. Then I adapted the preinstalled "QwertyDevice" and made a new emulator device template, the "OperaMiniLargeDevice" (OMLD), with the usable screen size of 1200x705 pixels. I omitted the usual control buttons of the standard emulators to have more space for the browser.

Here are two pictures of what the result looks like.

OperaMiniLargeDevice is now available in 4 variants for different screen resolutions. So if you want to test the solution yourself you should follow the next steps:

Requirements:
  • Get the Java SE Development Kit (JDK) from here.
  • Get the Sun Java Wireless Toolkit 2.5.2 for CLDC from here.
  • Get the "OperaMiniLargeDevice" for Sun Java Wireless Toolkit from here.
  • Get Opera Mini Browser 4.2 from here. Be sure to download both the JAD and the JAR to the same directory. The generic advanced version is recommended.
  • Internet access on the PC
Installation (done for Win XP):
  • Install the JDK.
  • Install the Wireless Toolkit.
  • Unzip your downloaded version of "OperaMiniLargeDevice" (simply extract it, a directory will be created automatically). Copy this directory to Path-to-WTk\wtklib\devices\ . The complete path to OMLD on my machine looks as follows: "C:\WTK2.5.2\wtklib\devices\OperaMiniLargeDevice1024x705".
  • Then choose "OperaMiniLargeDevice" as the default device, e.g. via "Start->All Programs->Sun WTK->Default Device Selection".
  • Then use "Start->All Programs->Sun WTK->Run MIDP Application" to start Opera Mini via its JAD file.
  • The start screen should appear and you should now be able to launch Opera Mini by pressing F2.

Remark:

It might be that you receive the following error on starting Opera Mini:

OTA server emulation started ...
HTTPS server emulation started ...
Error: Reason = 36

The most obvious reason for this error is that the "MIDlet-Jar-URL" in the JAD does not point to the right JAR file. Try to open the JAD with a text editor, locate the property "MIDlet-Jar-URL" and make sure that ONLY the filename of the JAR is left in this entry, e.g. MIDlet-Jar-URL: opera-mini-4.2.13918-advanced-de.jar . You must not use a complete URL to the JAR in the web. And do not forget to put the JAD and the JAR in the same directory.

Navigation:

You can set the link focus and scroll through a site with ARROW keys. The ENTER key selects an action, e.g. a text field. Texts can be entered with the normal keyboard keys. You can copy from and paste text into text fields by pressing STRG+c and STRG+v. The keys F1 and F2 control the menu actions of Opera Mini and the emulator, e.g. security questions for internet access. Since version 0.5 you can use your mouse to control Opera Mini and to click on links.

METHOD-2
I have been using Opera from the past two years and I feel it’s a great browser. I wanted to use Opera Mini in my cell phone but unfortunately for some reason it never worked. Today I came across an awesome article which allows you to run Opera Mini on your PC! Now the question is why would anyone use Opera Mini when there’s Opera browser for PC. Well, the answer is simple, if you have a slow internet connection like 56k internet connection or lesser then you’re benefited from this. Using Opera Mini you can load web pages way faster than any other browser.
Follow the below instructions carefully to use Opera Mini on your pc:
1. Download the microemulator from here and unzip the package to a specific folder.
2. Now download Opera Mini .jar and .jad files from here and save them in the same folder as it will be easy to browse.
3. Double click on “microemulator.jar” to run it. If it doesn’t start then you need to type “java -jar microemulator.jar” in a console.
4. Go to Options > Select device> Add…, now navigate to “microemulator/devices” and select “microemu-device-resizable.jar”.
5. Select the “Resizable device” profile you added and click on “Set as default”.
6. Click on the “Resize” button in the lower right corner and enter your preferred size.
7. Go to File> Open JAD and choose the Opera Mini .jad which you had downloaded in Step2.
8. “Opera Mini” will now appear in the list of applications you can start.
9. Click “Start” to launch it.

What is the obstacle to one-pointed meditation? The answer is the unstill mind. All problems are caused by the mind, by the desires arising in it. It is not easy to control the mind and keep it away effectively from desire. If we ask the mind to think of an object, it seems to obey us for a moment, but soon it takes its own course, wandering off. When I speak to you about meditation and tranquillity, for a moment your mind will perhaps become still and you will be happy. But in a trice it will go astray and the calm you experienced for a few seconds will give place to unquietness.
If you bid your mouth to keep shut, it obeys you for a brief moment. Similarly, if you close your eyes asking them not to see anything, they shut themselves off from the outside world for some moments. But try as your might to tell your mind not to think of anything, it will not listen to you.
The mind must be kept under control. Thinking and non-thinking must be govenred by your will. Only then can we claim that it is under our control, that we are masters of our consciousness.
Lunatics are usually referred to as people with no control over their minds. In fact none of us has any control over the mind. A mad man keeps blabbering. But what about us? We let the mind go freely to keep blabbering inwardly.
Do you know what it means to have mental control? Suppose you are suffering from a severe pain. If you ask your mind not to feel the pain, it shall not feel it in obediance to you (that is you will not feel the pain). Even if a tiger comes face to face with you and growls, you will feel no fear if you ask your mind not to be afraid of the beast. Now we keep crying for no reason. If the mind is under control we will keep smiling even if there is cause for much sorrow. And under the gravest of provacations it will not be roused to anger and will remain calm.
First we must train our mind not to keep wandering. One way of doing it is to apply it to good activities. When oil falls in a steady flow, without spraying, it is called "tailadhara". The mind must be gathered together and made steady. It must be accustomed to think of noble and exalted objects like the Lord. Eventually, the very act of "thinking" will cease and we will dissolve in Isvara (God) to become Isvara.
Before we pass on, we must find a way to control the mind. Otherwise, we will be born again and we will be subject to the constant unquietness of the mind again. So we must use the opportunity of this birth itself to subdue the mind even while we are in the midst of so much that can rouse our desire or anger. A man who has succeeded in bridling his mind thus is called a "yukta" by the yogins. He is a "sukhin", one who truly experiences bliss, so says Sri Krishna.
You must not turn away from yoga thinking that it is meant only for people like the sages. Who needs medicine? The sick. We suffer from manovyadhi, mental sickness. So we must take the medicine that cures it.
There are two different ways of mastering the mind - the first is outward (bahiranga) and the second is inward (antaranga). We must have recourse to both. The Matha has a cartman and a cook. Their work is outward in nature. Then there are those who prepare the wicks of the lamps, gather flowers for the pooja-they are "inward" workers. Both types are needed for the functioning of the Matha. By employing both the outward and inward means, the mind must first be applied to good things one-pointedly and eventually led to a state in which it does not think of anything at all.
The ourward means consists, for example, of sandhyavandana, sacrifices, charity and so on. The best inward means is meditation. There are five inward (or antanranga) means to aid meditation. They are ahimsa (non-violence), satya (truthfullness), asteyam (non-stealing), saucha (cleanliness) and indriya-nigraha (subduing the senses, if not obliterating them). To practise ahimsa is to imbue the mind with love for all and not even think of harming others. Asteyam means not coveting other people's goods. For satya, or truthfullness, to be complete one's entire being, including body, mind and speech, must be involved in its practise. Saucha is hygiene, observing cleanliness by bathing, maintaining ritual purity, etc. Indriya-nigraha implies limits placed on sensual enjoyment. "The eyes must not see certain things, the ears must not hear certain things and the mouth must not eat certain things" - restrictions with regard to what you can see, listen to, eat and do with your body. The body is meant for sadhana, for Atmic discipline. The senses must be "fed" only to the extent necessary to keep the body alive. These five dharmas are to be practised by all Hindus without any distinction of caste or community.

How to Fight Depression, Cheer Up Find Happiness

Positive Psychology Studies Uncover Secrets of Happiness


Happiness is a universal human goal. Aristotle recognized this over two-thousand years ago, when he said, "Happiness is the meaning and the purpose of life, the whole aim and end of human existence."
If generation after generation seeks greater happiness, why is it that depression is at an all time high in developed countries, and the onset of depression is occurring at younger and younger ages? Positive psychology research is bringing the tools for greater happiness within everyone's reach.

The Components Contributing to Happiness

Positive psychology studies identify three components contributing to an individual's happiness:
  • Set point
  • Circumstances
  • Intentional Activities.
Set point is a person's natural level of happiness; some people are just naturally happier than others. Research studies find that set point contributes about fifty-percent of a person's overall happiness. The set point of happiness derives from inherited genetics and childhood upbringing, and, therefore, is not within one's control.
Circumstances are the second component of happiness. Circumstances include a person's health, financial situation, relationship situations, and a wide array of other environmental factors over which a person has limited control. Psychology studies indicate about ten-percent of one's overall happiness derives from life's circumstances.
Intentional Activities are those things people do because the activity is pleasant and brings feelings of happiness. Since intentional activities are totally within a person's control, this is the area one should address to find greater happiness. Fortunately, intentional activities contribute about forty-percent of one's overall happiness, thereby providing a significant opportunity to find happiness through chosen activities.

The Secret of Happiness

Most people seek greater happiness by addressing their circumstances—such as trying to make more money, landing a better job, finding a better relationship, moving to a better part of town, or buying a new car.
Evaluating the three components of happiness—set point, circumstances, and activities—suggests that the secret of happiness is to address the single component one can control—intentional activities. Sadly, most people focus on circumstances, a happiness component over which they have little control, and more importantly, a component contributing only ten-percent to overall happiness.
The limited impact of circumstances on happiness seems counter-intuitive at first glance. Getting a raise or promotion, finding a new relationship, or buying something new, such as a car or new clothing, does make a person happier. However, this happiness is short-lived due to a phenomenon psychologists term hedonic adaptation, which means that humans tend to adjust rather quickly to life circumstances.
The new car brings a rush of good feelings for the first few days or weeks, but those feelings gradually fade as the proud owner adjusts and the new car becomes just a car. This phenomenon is observed with even significant changes in wealth. After a year, lottery winners return to their level of happiness before winning, having adjusted to the new level of wealth.

Intentional Activities that Bring Happiness

Positive psychology researcher, Sonja Lyubomirsky identifies twelve activities proven to bring greater happiness in her book, The How of Happiness: A Scientific Approach to Getting the Life You Want (Penguin Press, 2008). Dr. Lyubomirsky is Professor of Psychology at the University of California, Riverside with an undergraduate degree with honors from Harvard University and a Ph.D. from Stanford University.

Happiness Enhancing Strategies

Dr. Lyubomirsky's strategies involve ways of being, doing, and thinking that have been shown to increase happiness in controlled psychological studies. These strategies include:
  • Counting your blessings, expressing gratitude for what you have;
  • Cultivating optimism, learning to think optimistically;
  • Avoiding dwelling on problems and social comparisons;
  • Practicing acts of kindness;
  • Nurturing Relationships;
  • Doing more activities that truly engage you;
  • Savoring life’s joys;
  • Committing to goals;
  • Developing strategies for coping;
  • Learning to forgive;
  • Practicing religion and spirituality; and
  • Taking care of your body.

Take Control, Find Happiness Through Activities

Take control and find greater happiness by choosing things to do and ways of thinking that bring greater pleasure. The secret of happiness is to focus on what can be controlled and accept what cannot, choosing to engage in activities bringing lasting pleasure.
